Bill O'Reilly and Martin Dugard divide Killing the Mob: The Fight Against Organized Crime in America into three sections: “The Gunners,” “The Corruptors,” and “The Subversives.” Each part serves as a puzzle piece, completing for the readers the picture and history of organized crime in America and the names behind them.

O'Reilly and Dugard set the first part of the book in the 1930s, following the records of inglorious personalities who violently shook the foundation of American safety and security. The title "Public Enemy Number One" was popularized by the young lawyer John Edgar Hoover when he gave it to John Dillinger, the gang leader responsible for the robbery of several financial institutions and the murder of several innocent men. When asked about the most memorable moment of his career years later, Hoover chose the night they finally caught Dillinger. However, Dillinger's capture and subsequent death did not serve its purpose of warning big-time criminals—it was only the beginning of a long history of notorious syndicated transgression.

Soon, the names Bonnie Parker and Clyde Barrow, best known as the duo "Bonnie & Clyde," Charles Arthur "Pretty Boy" Floyd, George "Baby Face" Nelson, and Alvin Karpis made headlines with their respective criminal records, giving authorities a run for their confidence and pride. As it turned out, "Public Enemy Number One" is only a temporary label and needs to be passed on when the current titleholder is arrested. 

Meanwhile in Chicago  Alfonso "Al" Capone lords over as Public Enemy Number One. However, his type of crime was not as simple as robbing banks—they're more like a complex web of offenses that include extortion and murder for hire, among others. What makes Capone special, according to O'Reilly and Dugard, is that he does not operate informally. Instead, he was a member of a larger body of outlaws whose aim was to monopolize the criminal activities in various cities. They call themselves the Mafia, Black Hand, Cosa Nostra, the Organization, or simply, the Mob.

Hoover thought that the demise of his "Public Enemies" meant his triumph in the war on crime. However, he would soon learn the hard way that the Mob is far more dangerous than any gangster he arrested. Wiping out gangsters was enough to convince Hoover that crime was under control. But as the Second World War arrived in Europe, the Federal Bureau of Investigation (FBI) shifted its focus on enemy spies. While everyone focused on another set of law enforcement problems, the war was already breeding a new era of criminal organizations that would terrorize and corrupt the country. Borrowing from O'Reilly's and Dugard's words, "The Mafia is on its way."

The second part begins with Erwin Rommel, a German Field Marshal who anticipated a huge victory against the Allied troops under the command of British general Bernard Law Montgomery in North Africa. However, his plan was intercepted, leading his team, the Desert Fox, into a debacle—thanks to the Americans entering the battle fray. Rommel withdrew his army and flew to Eastern Europe, where he met the führer, Adolf Hitler, at the Werwolf, the latter's command post in Ukraine. The Third Reich lost thousands of men in the wake of the defeat and surrender of the German Afrika Korps on May 13, 1943.
