THE ROSIE PROJECT TELLS THE STORY OF DON TILLMAN, WHO LIVES IN Australia and has a job as a genetics professor. He has difficulty relating to other people, which has hindered him from having a fulfilling romantic relationship. Don finds it puzzling that he cannot find a woman to have a satisfying relationship with. He believes that his intelligence, health, income, and social status should all make him a highly qualified and appealing partner.
Don has two friends named Gene and Claudia, who are married. Eventually, after talking to Gene and Claudia, Don develops a questionnaire that will help him find the perfect mate for him. He calls this “The Wife Project.” Don soon finds out that his questionnaire is keeping women from being attracted to him, and he is unable to find an adequate woman.
In an attempt to help Don find a partner, Gene introduces him to Rosie. Rosie is immediately eliminated as a potential partner because she does not meet the criteria Don created in his questionnaire. However, Don is still drawn to Rosie and agrees to help her find the identity of her biological father. Rosie is unable to ask her mother who her biological father is because her mother has passed away. Don labels this quest to find Rosie's father “The Father Project.”
Don uses the laboratory at his university to DNA test the men who could potentially be Rosie's father. Many of them are eliminated immediately. Don and Rosie grow closer as they continue their search. Don is surprised to learn that Rosie is only working as a bartender to pay for her education and complete her Ph.D. in psychology. He learns that Rosie wants to find her biological father because her relationship with the man she knows as her father, Phil, is strained.
Don and Rosie must travel to New York City to test the DNA of two of the last remaining potential fathers of Rosie. Rosie convinces Don to break out of his routines and schedules while they are in New York, and to his surprise, Don enjoys the spontaneity. The two of them eventually realize they love each other. However, Rosie rejects Don because she is unsure that Don could love her in the way that other people can. Don explains that he can love but is unable to feel empathy.
When they return to Australia, Don realizes that Gene could be the father of Rosie. He decides to run a DNA test on Gene, which turns out to be negative. Don realizes that there is one person he has not run a DNA test on – Phil, who Rosie calls her father but does not believe is her biological father. Don runs the DNA test on Phil, and the test shows that Phil is actually Rosie's biological father. Rosie agrees to marry Don at the end of the novel, and the two of them return to New York City to start their lives over.
